TITLE:                    Enantioselective synthesis of alpha-quaternary amino acid derivatives. Application to alkaloid synthesis.

                                

ABSTRACT:           

 

 α-Quaternary amino acid derivatives are compounds that are used in the synthesis of modified peptides and proteins not found in nature. A combination of porcine liver esterase (PLE) catalyzed desymmetrization of α,α-disubstituted dimethyl malonates and the Curtius rearrangement was used in the enantioselective synthesis of a variety of α-quaternary amino acid derivatives that include N-Cbz protected amino esters, amines, carboxylic acids and ureas. The two-step methodology was then employed in studies toward the enantioselective synthesis of the alkaloid (-)-adalinine, a natural product isolated from the defence secretions of the lady bug, Adalia bipunctata.
