SACRI antibody services (SAS) has a dedicated protein purification facility and extensive experience with various purification techniques.  Our SAS staff also have considerable experience with advanced antibody applications requiring purified antibodies1.  Antibody purification can be used to concentrate and enrich antigen-specific antibodies, and remove unwanted proteins lowering background labeling in certain assays.  We work closely with clients to determine the best purification technique to obtain the highest yield and required purity for your application.  Our clients receive a detailed Account of Service compiled by our team of SAS purification specialists that includes a quality control report and an analysis of the increase in yield and activity.  In addition to implementing a protocol clients previously prepared, we can develop a custom purification protocol to suit your specific requirements.

Purification techniques

SAS uses an assortment of purification techniques including protein precipitation, gel filtration, and chromatography using our automated ÄKTA (GE Healthcare) fast protein liquid chromatography (FPLC) machine.  We can purify antibodies from serum, tissue culture supernatant, ascites, and other sources from any species.

Protein precipitation

SACRI antibody services offers a rapid protein precipitation service, employing both ammonium sulfate, sodium sulfate, and caprylic acid based techniques.  Coupled with other purification procedures, this is often the most cost-effective method to concentrate and purify large quantities of antibodies.  SAS can discuss the applicability of precipitation techniques to your needs.

Chromatography

With our new automated FPLC machine, SAS now offers a variety of chromatography techniques.  We can purify your antibody by selective affinity (e.g. Protein A, Protein G, etc.), size-exclusion, ionic exchange, hydrophobic interaction, and immunoaffinity.  Contact us to discuss your numerous options.

Quality control measures

Several quality control measures are available to ensure clients are supplied with the highest quality of purified proteins.  We will test the purity of your antibody at several stages during purification using SDS-PAGE electrophoresis, and will quantify the increased binding affinity if an antigen is available.
